ABSTRACT

Abstract Bamboo (Bambusa sp.) is a grass species with high potential for exploitation, however, this raw material is easily attacked by xylophagous agents such as Coleoptera. The objective of this study is thus to analyse the presence of wood-boring beetles associated with Bambusa sp. in Alta Floresta, Mato Grosso, Brazil. The study was carried out in a bamboo plantation, from August 2016 to July 2017. Two models of ethanolic traps were used, PET Santa Maria and ESALQ-84. Three families of wood-boring beetles were observed in this study: Bostrichidae, Cerambycidae and Curculionidae. 2,144 individuals were identified, belonging to 19 species, in four subfamilies and 11 tribes, and the species Xyleborus affinis was the most frequently collected. Thus, this study constitutes an important contribution to the knowledge of coleoborers associated with bamboo plantations, with the registration of the occurrence of species, as well as contributing to the knowledge of the entomofauna associated with species cultivated in the Southern Amazon.


Resumo O bambu (Bambusa sp.) é uma espécie de gramínea com alto potencial de exploração de fibras, no entanto, essa matéria-prima é facilmente atacada por agentes xilófagos, como os coleópteros. O objetivo deste estudo foi analisar a presença de besouros associados a Bambusa sp. em Alta Floresta, Mato Grosso, Brasil. O estudo foi realizado em um plantio de bambu, no período de agosto de 2016 a julho de 2017. Foram utilizados dois modelos de armadilhas etanólicas, o PET Santa Maria e ESALQ-84. Três famílias de coleobrocas foram observadas neste estudo: Bostrichidae, Cerambycidae e Curculionidae. Foram identificados 2,144 indivíduos, pertencentes a 19 espécies, em quatro subfamílias e 11 tribos, e Xyleborus affinis foi a espécie com maior número de indivíduos coletados. Assim, este estudo constitui-se em uma importante contribuição para o conhecimento de coleobrocas associadas a plantios de bamboo, com o registro de ocorrência de espécies, bem como contribui para o conhecimento da entomofauna associada a espécies cultivadas na Amazônia Meridional.

Abstract The cotton boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), is a key cotton crop pest in Brazil. Adverse climatic factors, such as high temperatures and low soil moisture, dehydrate oviposited cotton squares (bud flowers) on the ground and cause high mortality of its offspring within these plant structures. The objective of this research was to evaluate the mortality of the cotton boll weevil in drip and sprinkler irrigated cotton crops. The experimental design was in randomized blocks with two treatments: drip (T1) and sprinkler (T2, control) irrigated cotton crops with sixteen replications. Each parcel had one emergence cage, installed between two cotton rows per irrigation system, with 37 cotton squares with opened oviposition punctures and yellowish bracts, to capture adult cotton boll weevils. The average number of boll weevils that emerged from the cotton squares and the causes of mortality at different development stages were determined per treatment. Third-generation life tables of the boll weevil were prepared using the natural mortality data in drip and sprinkler irrigation treatments and plus actual, apparent and indispensable mortality rates and the lethality of each mortality cause. We conclude that the application of water directly to the root zone of the plants in a targeted manner, using the drip irrigation system, can cause high mortality of the cotton boll weevil immature stages inside cotton squares fallen on the ground. This is because the cotton squares fallen on the drier and hotter soil between the rows of drip-irrigated cotton dehydrates causing the boll weevils to die. This is important because it can reduce its population density of the pest and, consequently, the number of applications of chemical insecticides for its control. Thus, contributing to increase the viability of cotton production, mainly in areas of the Brazilian semiarid region where the cotton is cultivated in organic system.


Resumo O bicudo-do-algodoeiro, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), é uma praga-chave da cultura do algodão no Brasil. Fatores climáticos adversos, como altas temperaturas e baixa umidade do solo, desidratam os botões florais de algodão ovipositados caídos ao solo e causam alta mortalidade de seus descendentes dentro dessas estruturas vegetais.O objetivo desta pesquisa foi avaliar a mortalidade do bicudo-do-algodoeiro em lavouras de algodão irrigadas por gotejamento e aspersão. O delineamento experimental foi em blocos casualizados com dois tratamentos: cultivo de algodão irrigado por gotejamento (T1) e aspersão (T2, testemunha) com dezesseis repetições. Cada parcela possuía uma gaiola de emergência, instalada entre duas fileiras de algodão por sistema de irrigação, com 37 botões florais de algodão com orifícios de oviposição, brácteas abertas e amarelecidas, para captura do bicudo adulto. O número médio de bicudos que emergiu dos botões florais de algodão e as causas de mortalidade, em diferentes estágios de desenvolvimento, foram determinados por tratamento. As tabelas de vida de terceira geração do bicudo-do-algodoeiro foram preparadas usando os dados de mortalidade natural em tratamentos de irrigação por gotejamento e aspersão e as taxas de mortalidade reais, aparentes e indispensáveis e a letalidade de cada causa de mortalidade. Concluímos que a aplicação de água diretamente na zona radicular das plantas de forma direcionada, utilizando o sistema de irrigação por gotejamento, pode causar altas mortalidade dos estágios imaturos do bicudo-do-algodoeiro nos botões florais de algodão caídos no solo. Isso ocorre porque os botões florais de algodão caídos no solo mais seco e mais quente entre as fileiras do algodão irrigado por gotejamento desidratam, causando a morte dos bicudos. Isso é importante por poder reduzir a densidade populacional desse inseto e, consequentemente, o número de aplicações de inseticidas químicos para seu controle. Assim, contribuindo para aumentar a viabilidade da produção de algodão, principalmente em áreas do semiárido brasileiro em sistema orgânico.

ABSTRACT

Among the management and control tactics of the coffee berry borer Hypothenemus hampei (Ferrari), there is the use of entomopathogenic fungi. Due to the importance of prospecting isolates of entomopathogenic fungi for the control of the coffee berry borer, the objective of this study was to evaluate the efficiency of 26 isolates of entomopathogenic fungi in the control of this insect pest in the laboratory. The coffee berry borers were immersed in a solution adjusted to the concentration of 1 to 3 × 108 conidia/mL of each isolate and the control treatment (sterilized water). After seven days total mortality and confirmed mortality were evaluated. The isolates that caused the highest mortality and two commercial isolates were selected for evaluation of lethal concentration (LC50 and LC90) and lethal time (LT50 and LT90). Coffee berry borers were treated at different conidia concentrations for lethal concentration to assess total and confirmed mortality. For a lethal time, the coffee berry borers were treated at the concentration of 108 conidia/mL of the selected isolates, and, after two days and every 24 hours until the eighth day, the number of dead individuals was verified. Among the 26 isolates evaluated, 24 presented mortality higher than the control treatment, and three presented mortality higher than 85%. In the LC50 and LC90 assays, the IBCB 353 and IBCB 364 isolates were more lethal to H. hampei. In the LT50 and LT90 assays, the IBCB 66 and IBCB 353 isolates caused lethality in a shorter time.

Coffee is the most appreciated beverage in worldwide; Brazil is the largest producer and exporter of this commodity. Organochlorine endosulfan was banned from the country in 2013 due to its teratogenic agent-related features. Since then, coffee plantations have experienced increased Hypothenemus hampei infestation rates. The aim of the current study is to assess variations in the rates of Coffea arabica fruits brocaded by H. hampei after the application of entomopathogenic fungal species Beauveria bassiana IBCB66. Experiments were carried out with 'Catuaí' and 'Mundo Novo' cultivars between 2018 and 2020, during the borer transit period. Three experiments were carried out based on the application of the aforementioned fungal species on the investigated coffee plant species, both by spraying and sprinkling, at 30-day intervals; 10 fruits were collected per face of each useful plant in each repetition. The experiment has followed a randomized blocks design with five treatments, including the control, and five repetitions, each. Beauveria bassiana Ecobass (IBCB66) wettable powder spray, at the concentration of 2 × 1013·ha­1, was used in experiments I and II. On the other hand, the mix used in experiment III was prepared with blastospores at concentration of 5 × 1012·ha­1 blastospores + 0.1% Silwet. The sprinkling process in all three experiments has used dry aerial conidia at concentration of 2 × 1013·ha­1. Collected data were subjected to analysis of variance (ANOVA), which was followed by Fisher's test at 5% probability level, in the SISVAR software. More than 35,000 fruits were assessed. In addition to variations between experiments, results have evidenced that the rate of brocaded fruits remained high.

ABSTRACT

Endosymbiont bacteria can affect biological parameters and reduce the effectiveness of natural enemies in controlling the target insect. The objective of this work was to identify endosymbiont bacteria in Anaphes nitens (Girault, 1928) (Hymenoptera: Mymaridae), the main natural enemy used to manage Gonipterus platensis (Marelli, 1926) (Coleoptera: Curculionidae). Genomic DNA from six A. nitens populations was extracted and polymerase chain reactions (PCR) were performed with the primers to detect endosymbiont bacteria in this insect. The PCR products were amplified, sequenced, and compared with sequences deposited in the GenBank for the bacteria identification. All A. nitens populations had the bacterium Yersinia massiliensis (Enterobacteriales: Enterobacteriaceae). This bacterium was originally described as free-living, and it is associated with and composes part of the A. nitens microbiota. This is the first report of Y. massiliensis in an insect host.


As bactérias endossimbiontes podem afetar os parâmetros biológicos e reduzirem a eficácia de inimigos naturais no controle do inseto alvo. O objetivo deste trabalho foi identificar bactérias endossimbiontes em Anaphes nitens (Girault, 1928) (Hymenoptera: Mymaridae), o principal inimigo natural usado no manejo de Gonipterus platensis (Marelli, 1926) (Coleoptera: Curculionidae). O DNA genômico de seis populações de A. nitens foi extraído e as reações em cadeia da polimerase (PCR) realizadas com os primers para detectar bactérias endossimbiontes neste inseto. Os produtos de PCR foram amplificados, sequenciados e comparados com as sequências depositadas no GenBank para identificação das bactérias. Todas as populações de A. nitens tinham a bactéria Yersinia massiliensis (Enterobacteriales: Enterobacteriaceae). Esta bactéria foi originalmente descrita como de vida livre e está associada e compõe parte da microbiota de A. nitens. Este é o primeiro relato de Y. massiliensis em um hospedeiro.

Abstract The present study analyzed the volatile compounds emitted by Glycine max (cv. FT-Cristalina-RCH) soybean plants: healthy plants and plants damaged mechanically or by the Mexican soybean weevil Rhyssomatus nigerrimus. The SPME method was used to compare the volatile profile of soybean plants in four different conditions. The volatile profile of G. max plants infested by R. nigerrimus was qualitatively and quantitatively different from that of healthy and mechanically damaged plants. Emission of 59 compounds was detected in the four treatments. Of these compounds, 19 were identified by comparison of the Kovats index, mass spectrum and retention times with those of synthetic standards. An increase in concentration of the volatiles (Z)-3-hexenyl acetate and the compound 1-octen-3-ol was observed when the soybean plants were mechanically damaged. The compounds mostly produced by the soybean plant during infestation by male and female R. nigerrimus were 1-octen-3-ol, 6-methyl-5-hepten-2-one, (E)-β-ocimene, salicylaldehyde, unknown 10, linalool, methyl salicylate, (Z)-8-dodecenyl acetate (ester 5), ketone 2 and geranyl acetone. Behavioral effects of the identified compounds during the insect-plant interaction and their conspecifics are discussed.


Resumo O presente estudo analisou os compostos voláteis emitidos pelas plantas de soja Glycine max (cv. FT-Cristalina-RCH): plantas e plantas sadias danificadas mecanicamente ou pelo gorgulho da soja mexicana Rhyssomatus nigerrimus. O método SPME foi utilizado para comparar o perfil volátil de plantas de soja em quatro diferentes condições. O perfil volátil das plantas de G. max infestadas por R. nigerrimus foi qualitativa e quantitativamente diferente do das plantas saudáveis e danificadas mecanicamente. Foi detectada, nos quatro tratamentos, emissão de 59 compostos, dos quais 19 foram identificados por comparação do índice de Kovats, espectro de massa e tempos de retenção com aqueles de padrões sintéticos. Um aumento na concentração dos voláteis acetato de (Z)-3-hexil e do composto 1-octeno-3-ol foi observado quando as plantas de soja foram mecanicamente danificadas. Os compostos produzidos principalmente pela planta de soja durante a infestação por R. nigerrimus macho e fêmea foram 1-octeno-3-ol, 6-metil-5-hepteno-2-ona, (E)-β-ocimeno, salicilaldeído, desconhecido 10, linalol, salicilato de metila, acetato de (Z)-8-dodecenila (éster 5), cetona 2 e geranil acetona. Foram discutidos os efeitos comportamentais dos compostos identificados durante a interação inseto-planta e seus coespecíficos.

Abstract Rhynchophorus ferrugineus (RPW) (Olivier) (Coleoptera: Curculionidae) is one of the most destructive pest of palm according to dozens of literature, a lot of effort have been made since three decades up to date to solve this problem, one of newest solution raised is using nano pesticides. Imidacloprid and Chloropyrophos and their nano form were tested against 10 days-old larvae and newly emerged adults of the red palm weevil. The pesticides had toxicological and pathological effects on Rh. ferrugineus larvae and adults. The toxicity effect of nano chloropyrophos was more than others. The damage included vaculation of cytoplasm, analyzes and destroyed nuclei of the epithelial cells. The larvae were more sensitive in the total damages in comparison with adults. This investigation is the first record to the histopathological effects of nano pesticides.


Resumo Rhynchophorus ferrugineus (RPW) (Olivier) (Coleoptera: Curculionidae) é uma das pragas mais destrutivas da palmeira, de acordo com dezenas de publicações, muito esforço foi feito desde três décadas para resolver esse problema, uma das mais recentes soluções levantados está usando nano pesticidas. O imidaclopride e o cloropirofós e sua nano forma foram testados em larvas com 10 dias de idade e em adultos recém-emergidos do gorgulho da palma vermelha. Os pesticidas tiveram efeitos toxicológicos e patológicos em Rh. larvas de ferrugineus e adultos. O efeito da toxicidade dos nano cloropirofos foi maior que outros. O dano incluiu a vacinação do citoplasma, análises e núcleos destruídos das células epiteliais. As larvas foram mais sensíveis no total de danos em comparação com os adultos. Esta investigação é o primeiro registro dos efeitos histopatológicos dos nano pesticidas.

ABSTRACT

Bark and ambrosia beetles, mainly the ones belonging to groups Scolytinae, Bostrichidae and Platypodinae, can kill trees from reforestation areas or native forests and damage the wood. Population monitoring and the identification of quarantine species are carried out by assembling ethanol-baited traps. The aims of the current study are to evaluate the influence of the color of ethanol-baited traps on the efficient capture of these insects, as well as to measure changes in colorimetric variables based on trap exposure in the field and to investigate whether these changes affect capture efficiency. Eight ethanolic traps (red, yellow, black and transparent traps - two of each color) were installed in a forest fragment in the first experimental stage ­ samples were collected on a weekly basis, for 11 months. New and used transparent traps were installed in the field in the second experimental stage - samples were collected for additional 11 months. A portable spectrophotometer was used to measure the colorimetric variables in these traps. The mean number of Scolytinae individuals (± SD) captured in transparent traps (48±50) was significantly higher than that of individuals captured in black (24±25), yellow (23±21) and red (22±21) traps. However, transparent traps subjected to field conditions were colonized by such as fungi, bacteria and mosses, which changed the transparent state of the traps into a darkened color and significantly affected their capture efficiency. The total number of 6,268 Scolytinae individuals were collected at this experimental stage: 4,977 of them were captured in new traps, whereas 1,291 were captured in the old ones. Based on the herein measured colorimetric variables, such color change got significantly intensified as transparent traps remained under field conditions. In conclusion, transparent traps were more efficient in capturing Scolytinae individuals than the black, yellow and red traps. In addition, the exposure to field conditions has progressively changed equipment color and decreased its capture efficiency.

ABSTRACT

Weevil is one of the main pests of stored corn. The adults feed on intact and broken seeds or bran, while the larvae develop by feeding on the inside of the seeds, resulting in a reduction in weight and physiological quality. The objective of this study was to evaluate the effects of Sitophilus zeamais infestation on corn seeds during storage. Corn seeds were infested using insect densities: 0, 36, 72, 108, and 144 individuals per 240 g of seeds. The infested seeds were placed in individualized plastic containers and covered with voile fabric. The seeds were stored for 60 days under laboratory conditions with an average temperature of around 27 °C and relative humidity of 85%. Initially and every 15 days of storage for 60 days, the seeds were evaluated using the parameters: number of S. zeamais individuals, total number of infested seeds, seed weight loss, germination test and seedling emergence in the tray. The population increase of S. zeamais causes a decrease on the physical and physiological quality of stored corn seeds, but the deterioration rate does not increase in the same proportion as the weevil population growth rate.

ABSTRACT

Sweet potato, Ipomoea batatas (L.) Lam (Convolvulaceae), is an essential crop for food security in developing countries. Euscepes postfasciatus (Fairmaire) (Coleoptera: Curculionidae) is one of the main pests of I. batatas in tropical and subtropical regions. It feeds on the tuberous roots of I. batatas and induces perforations tunnel-shaped with excrement. The objective of this study was to register, for the first time, the occurrence of E. postfasciatus in I. batatas in the municipality of Diamantina, Minas Gerais, Brazil. Individuals of E. postfasciatus were found in the larval, pupal and adult stages feeding on the roots, in the genotypes, Brasilândia Branca, Rubisol, UFVJM01, UFVJM02, UFVJM03, UFVJM04, UFVJM08, UFVJM18, UFVJM91, UFVJM291 and UFVJM526 in a greenhouse at the Universidade Federal dos Vales do Jequitinhonha e Mucuri (UFVJM). Euscepes postfasciatus injuries formed superficial and deep galleries with the presence of excrement and unpleasant odor. The record of E. postfasciatus in I. batatas in the municipality of Diamantina is important to develop local strategies for integrated pest management of the crop in the region.(AU)

Poor storage conditions provide favorable environment to stored grain pests for their growth. The bio-pesticides are the best alternatives to synthetic pesticides. Present study was conducted to compare toxicity of Rubus fruticosus and Valeriana jatamansi against granary weevil, Sitophilus granarius and subsequent changes in enzyme activity responsible for grain damage. In current research 5 g of R. fruticosus fruit and V. jatamansi rhizome powders were tested separately against S. granarius, in 50 g wheat whole grains for seven days in comparison with the control. The enzymatic activity of malate dehydrogenase and α-amylase was observed in the cellular extracts of S. granarius. The insects were crushed and homogenized in phosphate-buffer solution and centrifuged at 10000 rpm for 5 minutes. For the enzymatic measurement supernatant was tested; the spectrophotometer was adjusted at 340 nm. The reagents were mixed and incubated at 25 °C for five minutes. The cuvettes were placed in the experimental and reference sites of spectrophotometer and recorded the change in absorbance for 3-4 minutes. There was 5.60% and 14.92% reduction in the activity of malate dehydrogenase in R. fruticosus and V. jatamansi, treated insects, respectively. The alpha amylase enzyme activity was 6.82% reduced and 63.63% increase in R. fruticosus and V. jatamansi, treated insects, respectively. Present study addresses that both plant powders are effective against granary weevil by altering enzyme activities so both the plant powders can be used as bio-pesticides against the stored grains pests.


As más condições de armazenamento proporcionam um ambiente favorável às pragas armazenadas para o crescimento. Os biopesticidas são as melhores alternativas aos pesticidas sintéticos. O presente estudo foi conduzido para comparar a toxicidade de Rubus fruticosus e Valeriana jatamansi contra gorgulhos, Sitophilus granarius e subsequentes alterações na atividade enzimática responsáveis ​​por danos aos grãos. Na pesquisa atual, 5 g de frutos de R. fruticosus e pós de rizoma de V. jatamansi foram testados separadamente contra S. granarius, em 50 g de grãos integrais de trigo por sete dias, em comparação com o controle. A atividade enzimática da malato desidrogenase e α-amilase foi observada nos extratos celulares de S. granarius. Houve redução de 5,60% e 14,92% na atividade da malato desidrogenase em R. fruticosus e V. jatamansi, insetos tratados, respectivamente. A atividade da enzima alfa amilase foi reduzida em 6,82% e aumento de 63,63% em R. fruticosus e V. jatamansi, insetos tratados, respectivamente. O presente estudo aborda que ambos os pós de plantas são eficazes contra o gorgulho do celeiro, alterando as atividades enzimáticas, de modo que ambos os pós de plantas possam ser usados ​​como biopesticidas contra pragas de grãos armazenados.

Field experiments were carried out in the reclaimed land east of Cairo-Alexandria Desert Road, Village of Wardan, Giza Governorate, Egypt, to evaluate the impact of using nano-gels pheromone for the first time on the attraction of the RPW adults by pheromone traps compared to the control which is normal pheromone during two successive seasons in 2018 and 2019. Also, this study aimed to compare the efficacy of two types of trap, traditional trap (type 1) and dry funnel trap (type 2) in capturing of RPW adults. Results revealed that the nano gel pheromone traps attracted significantly more adults with an average of (4.26 and 3.56) adults/trap and total of (55.33 and 46.33) adults/trap compared to control with an average (2.69 and 2.46) and total of (35.00 and 32.00) adults/trap for the two seasons 2018 and 2019 respectively. Results indicated that the use of nano gel pheromone for the first time in field improved the trap catchability to RPW adults by 22.51 and 18.30% of total number of RPW adults captured throw out the two successive seasons respectively. In addition, there was no significant difference between the total numbers of RPW adults collected by the two tested type of traps over the two successive seasons. Furthermore, the sex ratio between males and females of total collected RPW adults found to be1: 1.43 and 1: 1.94 at 2018 and 2019 respectively.


Experimentos de campo foram realizados na terra recuperada a leste da estrada do deserto de Cairo-Alexandria, vila de Wardan, província de Giza, Egito, para avaliar o impacto do uso de feromônio de nano géis pela primeira vez na atração de adultos RPW por armadilhas de feromônio comparado ao controle que é o feromônio normal durante duas temporadas sucessivas em 2018 e 2019. Além disso, este estudo teve como objetivo comparar a eficácia de dois tipos de armadilha, armadilha tradicional (tipo 1) e armadilha de funil seco (tipo 2) na captura de RPW adultos. Os resultados revelaram que as armadilhas de nano gel de feromônio atraíram significativamente mais adultos, com uma média de (4,26 e 3,56) adultos/armadilha e total de (55,33 e 46,33) adultos/armadilha em comparação ao controle com uma média (2,69 e 2,46) e total de (35.00 e 32.00) adultos/armadilha para as duas temporadas 2018 e 2019, respectivamente. Os resultados indicaram que o uso de feromônio nano gel pela primeira vez em campo melhorou a capturabilidade de armadilhas para adultos com RPW em 22,51 e 18,30% do número total de adultos com RPW capturados durante as duas estações sucessivas, respectivamente. Além disso, não houve diferença significativa entre o número total de adultos RPW coletados pelos dois tipos de armadilhas testados ao longo das duas estações sucessivas. Além disso, a proporção sexual entre homens e mulheres do total de adultos coletados de RPW foi de 1: 1,43 e 1: 1,94 em 2018 e 2019, respectivamente.

ABSTRACT

We report the first record of occurrence of Loncophorus longinasus in fruits of munguba, Pseudobombax munguba, in the state of Amazonas, Brazil. Some morphological and biological aspects of L. longinasus are presented. Adults were obtained through natural emergence from munguba fruits that opened for seed dispersal. The fruits had perforations leading to galleries through the exocarp. Inside the fruit, larvae were found at the seed insertion points on the central seed attachment structure. The pupae developed in a cocoon at the same structure and turned mature simultaneously with the capsule opening. Together with the larvae, the presence of ants (Pseudomyrmex sp.) was observed, that apparently maintained the galleries open for access and feeding on larval exudates. (AU)

ABSTRACT

Five Zea mays cultivars (BRS Caatingueiro, BRS Gorutuba, BRS Sertanejo, BRS Asa Branca and BR 106) were evaluated considering their effect on the nutrition of the maize weevil Sitophilus zeamais, by analysis of total protein in adult fed with these cultivars and for the presence of lectins and trypsin inhibitors in grains. In addition, free-choice and no-choice assays were performed to investigate the resistance of grains of the Z. mays cultivars to an attack by S. zeamais. The BR 106 cultivar showed the lowest susceptibility index, followed by BRS Caatingueiro, BRS Asa Branca, BRS Sertanejo and BRS Gorutuba. The number of emerged adults in the Z. mays cultivars ranged from 213.17 to 74.0, and the lowest number of insects was recorded for the BR 106 cultivar. The insects were able to feed on grains of all cultivars, but the BR 106 cultivar showed the least reduction in dried biomass. Lectins were detected in extracts from BR 106, BRS Asa Branca, BRS Sertanejo and BRS Gorutuba, and the highest activity was shown by BR 106. The lowest protein assimilation was detected in the insects from treatments with BRS Asa Branca. The extracts from all cultivars were able to inhibit the activity of bovine trypsin, but this effect was not related to the resistance degree of Z. mays cultivars. The results suggest the resistance of BR 160 to the attack of S. zeamais, as well as indicating that the presence of lectin in the grains is the cause of this resistance.(AU)


Foram avaliadas cinco cultivares de Zea mays (BRS Caatingueiro, BRS Gorutuba, BRS Sertanejo, BRS Asa Branca e BR 106) e seu efeito na nutrição do gorgulho-do-milho Sitophilus zeamais, por meio da análise de proteína total em adultos alimentados com esses cultivares e a presença de lectinas e inibidores da tripsina nos grãos. Além disso, foram realizados ensaios com e sem chance de escolha para investigar a resistência dos cultivares de Z. mays ao ataque de S. zeamais. O cultivar BR 106 apresentou o menor índice de susceptibilidade, seguido por BRS Caatingueiro, BRS Asa Branca, BRS Sertanejo e BRS Gorutuba. O número de adultos emergidos nos cultivares de Z. mays variou de 213,17 a 74,0, e o menor número de insetos foi registrado para o cultivar BR 106. Os insetos foram capazes de se alimentar de todos os cultivares, no entanto, o BR 106 mostrou a menor redução na biomassa seca. As lectinas foram detectadas em extratos de BR 106, BRS Asa Branca, BRS Sertanejo e BRS Gorutuba, e a maior atividade foi demonstrada pela BR 106. A menor assimilação de proteína foi detectada nos insetos que se alimentaram com BRS Asa Branca. Os extratos de todos os cultivares foram capazes de inibir a atividade da tripsina bovina, mas esse efeito não está correlacionado ao grau de resistência dos cultivares de Z. mays. ABSTRACT

The weevil Ozopherus muricatus Pascoe, 1872 (Coleoptera: Curculionidae) is already considered a pest in Brazil for açaí palm (Euterpe oleracea Mart.) plantations. Its presence has become more frequent and caused significant reduction in the production of açaí fruit. Studies on the biology of this insect pest are still scarce, hindering its management. For studies on pheromones, for instance, it is essential to correctly identify the sex of the insect. The objective of this study was to evaluate the existence of sexual dimorphism in pupae and adults of O. muricatus in order to allow the identification of males and females. Larvae, pupae and adults were collected from açaí plantations and kept in biochemical oxygen demand (BOD) type incubation chambers. Adults and pupae were examined under a stereomicroscope, and dimorphism was determined through observation of external morphological characteristics. In addition, adults were dissected in order to observe genitalia and confirm the sex of specimens. In the pupae, sexual dimorphism was characterized by the presence of two protuberances in the terminal ventral region of the abdomen of females, which were absent in males. In adults, sexual dimorphism was also observed, based on the final abdominal segments. In males, it was possible to see all eight abdominal tergites, while in females tergite VIII is covered by tergite VII. Thus, the sexual dimorphism present in the morphological characteristics of O. muricatus allows the distinction between males and females of this species.(AU)


O gorgulho Ozopherus muricatus Pascoe, 1872 (Coleoptera: Curculionidae) já é considerado uma praga em plantações de açaí (Euterpe oleracea Mart.) no Brasil. Sua presença tem se tornado mais frequente e causado redução significativa na produção de frutos. Estudos sobre a biologia dessa praga ainda são escassos, dificultando o seu manejo. Para estudos com feromônios, por exemplo, é essencial a correta identificação do sexo do inseto. O objetivo deste estudo foi avaliar a existência de dimorfismo sexual em pupas e em adultos de O. muricatus, a fim de permitir a identificação de machos e fêmeas. Larvas, pupas e adultos foram coletados de plantações de açaí e mantidos em câmaras incubadoras para demanda bioquímica de oxigênio (DBO). Adultos e pupas foram examinados sob microscópio estereoscópico, e o dimorfismo foi determinado por meio da observação de características morfológicas externas. Adicionalmente, adultos foram dissecados para a observação da genitália e confirmação do sexo dos espécimes. Nas pupas, o dimorfismo sexual foi caracterizado pela presença de duas protuberâncias na região terminal ventral do abdome das fêmeas, as quais foram ausentes nos machos. Nos adultos, também se observou dimorfismo sexual, com base nos segmentos finais do abdome. Nos machos, é possível visualizar todos os oito tergitos abdominais, enquanto nas fêmeas o tergito VIII é encoberto pelo tergito VII. Assim, o dimorfismo sexual presente nas características morfológicas de O. muricatus permite distinção entre machos e fêmeas dessa espécie.(AU)

ABSTRACT

The chemical constituents and insecticidal activity of essential oil obtained by hydrodistillation of the leaves of Paullinia pinnata Linn (Sapindaceae) are being reported. The essential oil were analysed by using gas chromatography (GC) and gas chromatography coupled with mass spectrometry (GC-MS). Different concentrations (50, 100, 150, 200 and 250 mg/mL) of P. pinnata essential oil prepared separately and diluted in dimethyl sulfoxide (DMSO) were tested on the maize weevil, Sitophilus zeamais. The main constituents of the oil were pentadecanoic acid (17.9%), isoaromadendrene epoxide (11.5%) and wine lactone (11.2%). Other significant compounds of the essential oil were eremophilene (6.9%) and phytol (6.2%). The essential oil displayed 100% mortality (fumigant toxicity) against S. zeamais adults at tested concentration of 150 mg/mL with lethal concentrations (LC50) of 51.87 mg/mL air. This is the first report on the chemical constituents and insecticidal activity of essential oil of P. pinnata and may be explore as a potential natural herbal plant for the control of insect pest.


Se informan los componentes químicos y la actividad insecticida del aceite esencial obtenido por hidrodestilación de las hojas de Paullinia pinnata Linn (Sapindaceae). El aceite esencial se analizó mediante cromatografía de gases (GC) y cromatografía de gases acoplada con espectrometría de masas (GC-MS). Se ensayaron diferentes concentraciones (50, 100, 150, 200 y 250 mg/ml) de aceite esencial de P. pinnata preparado separadamente y diluido en dimetilsulfóxido (DMSO) en el gorgojo de maíz, Sitophilus zeamais. Los componentes principales del aceite fueron ácido pentadecanoico (17,9%), isoaromadendreno epóxido (11,5%) y vino lactona (11,2%). Otros compuestos significativos del aceite esencial fueron eremophilene (6,9%) y phytol (6,2%). El aceite esencial mostró una mortalidad del 100% (toxicidad fumigante) contra los adultos de S. zeamais a una concentración de 150 mg/ml con concentraciones letales (CL50) de 51,87 mg/ml de aire. Este es el primer informe sobre la composición del aceite esencial de P. pinnata y su actividad insecticida. Puede ser explorado como una potencial planta herbácea natural para el control de la plaga de insectos.

Preliminary bioassays with whole cultures (WC) of 124 Bacillus thuringiensis strains were performed with neonate larvae of Anthonomus grandis, a major cotton pest in Argentina and other regions of the Americas. Three exotic and four native strains were selected for causing more than 50% mortality. All of them were β-exotoxin producers. The native strains shared similar morphology of parasporal crystals, similar protein pattern and identical insecticidal gene profiles. These features resembled Lepidoptera-toxic strains. Furthermore, these strains showed a Rep-PCR pattern identical to lepidoptericidal strain HD-1, suggesting that these strains may belong to serovar kurstaki. However, some differences were observed in the plasmid profiles and in the production of β-exotoxin. To determine the culture fractions where the insecticidal metabolites were present, bioassays including resuspended spore-crystal pellets, filtered supernatants (FS) were compared with those of WC. Both fractions tested showed some level of insecticidal activity. The results may suggest that the main toxic factors can be found in FS and could be directly correlated with the presence of β-exotoxin. Based on the bioassays with FS and autoclaved FS, the participation of thermolabile virulence factors such as Cry1I in toxicity is neither discarded. In the selected strainsβ-exotoxin would be the major associated virulence factor; therefore, their use in biological control of A. grandis should be restricted. Nevertheless, these strains could be the source of genes (e.g., crylla) to produce transgenic cotton plants resistant to this pest.


Se realizaron ensayos preliminares con cultivos completos de 124 cepas de Bacillus thuringiensis utilizando larvas neonatas de Anthonomus grandis, una plaga principal del algodón en Argentina y otras regiones de América. Se seleccionaron 3 cepas exóticas y 4 nativas por producir mortalidad superior al 50%, todas ellas productoras de β-exotoxina. Las cepas nativas presentan la misma morfología de cristales, un perfil de proteínas similar y los mismos genes insecticidas. Estas características hacen que se parezcan a cepas tóxicas para lepidópteros. Además, mostraron un perfil de Rep-PCR idéntico al de la cepa lepidoptericida HD-1, lo que indica que podrían pertenecer al serovar kurstaki. Sin embargo, se observaron diferencias en el perfil plasmídico y en la producción de β-exotoxina. Para determinar en qué fracción del cultivo se encontraban los metabolitos responsables de la toxicidad, se compararon los resultados de bioensayos en los que se utilizó biomasa, sobrenadante filtrado (SF) o cultivos completos. Ambas fracciones mostraron cierto grado de toxicidad. Los resultados indican que los principales factores tóxicos se encuentran en el sobrenadante y estarían directamente relacionados con la presencia de β-exotoxina. De acuerdo con los bioensayos de SF y SF autoclavado, no se descarta también la participación en la toxicidad de factores de virulencia termolábiles, como CrylIa. En las cepas seleccionadas, el principal factor de virulencia es la β-exotoxina, por lo que su uso debería restringirse para el control biológico de A. grandis. No obstante, estas podrían ser fuente de genes (p. ej., crylIa) para la producción de plantas de algodón transgénicas resistentes a dicha plaga.
